Looking for a birth index entry in the 1960s, I noticed in passing a
couple of entries (for different people) had what looked like a
handwritten 2 in the left margin.
Is this chance, or does it have a significance?
(E.g. the second entry on a page in the third quarter 1967. To get to
the page with Ancestry, look for Bott in that quarter, which gets one
to page 78, and then move to page 79.]

It's the way in which additional (missed) entries are made to the GRO
indexes after they have been published. For example, the first
additional entry would have a number 1 placed close to the point at
which the entry should have appeared, and the details placed at the
bottom of the sheet. Further additional entries would be numbered 2,
3 etc. It's not very clear in the examples you have quoted as the
left hand margin has been truncated.
It's the additional entries on the right hand side of the page that
has screwed up the Ancestry indexing.

If I'd seen a 1 above the 2 (I hadn't thought it might be under the
paper blanking thev left nmargin), I might have guessed (or if
downloading on my machine was a bit faster, I might have seen the
insertions at the bottom of the page.)
Thanks for the answer to my question - and to the one I hadn't asked
but had wondered about.
